When used as instructed, these vessels are remarkably robust and can handle typical laboratory conditions, without fracturing or resulting in loss of volume. It is recommended that, when applying caps, a proper tray be used to protect the bottom of the tubes from excessive force that is typically used.

We recommend low profile PCR plates and tubes for reaction volumes below 20 µL. The low profile design minimizes the airspace between the PCR reaction and reduces evaporation effects, thereby increasing PCR efficiency in low volume reactions.

Answer:

Low profile PCR plastics have shorter wells than the standard profile plastics, decreasing the “dead space” between the heated lid of the thermal cycler and the sample. This eliminates condensation on the side wall of the tube, preventing reduction in PCR volume and increasing the efficiency of the reaction.

Answer:

PCR plastics are not typically sterilized as their entire production process, from molding to final packaging, is carried out in a Class 100,000 clean room under ISO 9001 guidelines. All PCR plastics are certified free from RNase, DNase and human DNA. In contrast, during typical non-clean room production, plastics are exposed to many contaminants including dust, bacterial cells, and DNA. The plastics are then sterilized to kill bacteria and inactivate RNases and DNases, but sterilization does not remove dust or DNA contamination. The dust particles left behind can inhibit PCR, and the damaged DNA fragments can still act as templates, leading to non-specific amplification.

How can I minimize evaporation loss?

The smaller the sample volumes, the more important it becomes to prevent evaporation loss. Thanks to the large selection of different sealing films and strips, you always have a perfectly harmonized system to securely seal your PCR plates and vessels.

Answer:

Although PCR consumables are made of PP (which in general is autoclavable at 121°C (2 bar, acc. to DIN EN 285) we do not recomment to autoclave these tubes. Mainly, because PCR products have very thin walls and secondly because the autoclave could be a source for contamination.

PCR sealing mats have been designed to fit PCR plates. They can reduce evaporation losses up to 75% compared to conventional systems. They are charcterized by their high flexibility, tight sealing, autoclavability and are easily pierced by pipette tips.

Answer:

Sealing mats can be autoclaved at +121°C (2 bar, acc. to DIN EN 285). However the silicone sealing mat (Cat. No. 781408) can only be autoclaved ~ 5 x because of material shrinkage.
